DISCOVERY OF NOVEL EPIGENETIC BIOMARKERS IN ORAL MALIGNANT LESIONS BY EPIGENOMICS AND TRANSCRIPTOMICS APPROACHES

چکیده

Objectives: DNA methylation, which is the most frequently observed epigenetic change, very important because it seen in early stages of cancer. Abnormal methylation promoter regions and silencing tumor suppressor genes play a key role development oral squamous cell carcinoma (OSCC). In our study*, was aimed to identify validate new biomarker candidates via epigenomics/transcriptomics approaches, understand molecular mechanisms OSCC discover biomarkers for diagnosis. Materials Methods: After DNA/RNA isolation tumor/matched-normal tissue samples from 6 patients, BC-DNA/cDNA synthesis performed, respectively. The expression profiles were analyzed by R(v3.5.1) environment methods using IlluminaHumanMethylation450chips IlluminaiScan, A candidate gene showing methylation-dependent loss after bioinformatic analysis validated QRT-PCR AND QMSP tissues body fluids 20 premalignant lesions (OPML) Results: To candidate, we selected show either hypermethylation lower patterns. This (unpublished data), belong subfamily protein-tyrosine kinase, found be methylated 65% tumors, 20% matched-normal OSCC. rates tumor, saliva OPML patients 55%, 40% 10%, decreased levels 55% 35% Conclusions: Our could detection OSCC.

ASSESSMENT OF TUMOR MARKERS IN BENIGN, DYSPLASTIC AND MALIGNANT ORAL MUCOSAL LESIONS

Immunohistochemical demonstration of tumor-associated markers may be valuable as an adjunct to conventional histology for determination of malignancy development in oral mucosal diseases. Accordingly, the present study determined the distribution of some important tumor markers such as heat shock proteins (HSP), fibroblast growth factors (FGF) and transferrin receptors (TF-R) in 22 samples ...

Differentiation of benign and malignant breast lesions by apparent diffusion coefficient value in mass and non-mass lesions

Background: Diffusion-weighted imaging (DWI) is one of methods in evaluation of breast lesions. We aimed to investigate the apparent diffusion coefficient (ADC) values in breast tumors and their accuracy in differentiating benign versus malignant lesions. Methods: In this cross-sectional study, 72 patients with 88 breast lesions were investigated by 1.5-T breast MRI from 2015 to 2017 in Athari...

Tissue eosinophils as a histopathological marker in oral premalignant and malignant lesions: a histochemical study

Introduction: Although recent studies show the association of eosinophils with different tumors including oral malignancies, the role of eosinophils in premalignant conditions of the oral cavity is not known precisely. Therefore, this study aimed to determine the number of eosinophils in premalignant and malignant conditions of the oral cavity and its role in progression to malignancy. Materia...

Discovery of novel biomarkers in oral submucous fibrosis by microarray analysis.

Oral submucous fibrosis (OSF) is a high-risk precancerous condition of the oral cavity. Areca nut chewing is its key etiologic factor, but the full pathogenesis is still obscure. In this study, microarray analysis was used to characterize the mRNA changes of 14,500 genes in four OSF and four normal buccal mucosa samples to identify novel biomarkers of OSF. Five candidate genes with the most dif...

Discovery of new biomarkers for malignant mesothelioma

Malignant mesothelioma is an asbestos-induced, aggressive tumour with limited treatment options and very poor outcome. Currently, there are no tumour biomarkers in widespread clinical use for this disease. Soluble mesothelin is the most intensively investigated mesothelioma biomarker and has been approved by the US FDA primarily as a tool for monitoring patient response and progression. Mesothe...
